What is HPLC testing?

High-performance liquid chromatography (HPLC) is the standard analytical technique for verifying research peptide purity. A dissolved sample is pushed through a chromatographic column; the resulting UV absorbance trace shows the target peptide as a peak alongside any impurities.

How HPLC purity is reported

Purity is expressed as a percentage of total peak area at a specified UV wavelength (typically 214 or 220 nm). Research-grade peptides target ≥99%.

Analytical vs preparative HPLC

Preparative HPLC purifies bulk peptide during manufacturing. Analytical HPLC is the QC step that certifies the finished batch — the trace behind the Certificate of Analysis.

What HPLC does not tell you

HPLC confirms purity but not identity. That is why we pair it with mass spectrometry — see Understanding COAs for peptides and Peptide purity standards.

Frequently asked questions

What does HPLC measure?

Purity by peak area at a specified UV wavelength — typically 214 or 220 nm for peptides.

Does HPLC confirm the peptide identity?

No. HPLC verifies purity; identity is confirmed by mass spectrometry.
